Giambattista Orsini was the son of Lorenzo orsini, Lord of Monterotondo (near Rome), and Clarice Orsini, sister of the powerful Latino Orsini. He was an important member of one of the the most powerful aristocratic families in Rome
. His uncle, Latino Orsini, was a Cardinal and Archbishop of Bari, and another uncle, Giovanni, was Abbot of Farfa.

Giambattista was made a Cardinal by Pope Sixtus IV in 1483. He went on the become Cleric of the Apostlic Chamber, Auditor of the sacred Rota, and Apostolic Protonotary. Pope Innocent VIII made him Archbishop of Taranto. Pope Alexander VI made him Papal Legate (Administrator) of Bologna, the Marche and the Romagna.

In the conclave of 1492, he was persuaded to support Rodrigo Borgia by the grant of benefices and the fortified towns of Monticelli and Soriano. Over time, the Orsini came to fear the growing power of Cesare Borgia and the pope. He left his legation of Marche Anconitana on July 29, 1500 and was named legate in Bologna. On July 12, 1502, he asked for an audience with the pope to request permission to leave for Milan to meet the king of France; he did not obtain permission but still left the following evening. He attended, in the proximity of the lake of Trasimène, a reunion of the chiefs of bands of the Orsinis conspiring against Cesare Borgia; no agreement was reached among them.

He returned to Rome and was called to the Vatican by Pope Alexander VI on January 3, 1503. He was arrested by orders of the pope, who had until then favored him in everything. The cardinal was taken to Tor di Nona, and later to Castel Sant'Angelo. His palace and all his possessions were confiscated; all the places owned by the Orsinis were delivered to Cesare Borgia. He fell ill in prison and died after twelve days on 22 February 1503.
